Because generation sequencing (NGS) to detect somatic mutations have been adopted in the clinical field, both qualitative and quantitative QC of somatic variants through the entire coding region was detected by NGS are very important. However, application or specific guidelines, especially for quantitative QC, currently insufficient.
Our goal is to design a practical approach to both quantitative and qualitative QC using the example of detecting clonal hematopoiesis indefinite potential (CHIP) .We implement a QC scheme using commercial materials and in-house reference materials QC (IQCM) consists of a map of haplotypes and cell lines cancer to monitor CHIP.This efficient approach validated assay adapted CHIP NGS.
Accuracy, analytical sensitivity, analytical specificity, precision qualitative (concordance), and the detection limits were reached 99.87%, 98.53%, 100.00%, 100.00%, and 1.00%, respectively. Analysis of quantitative precision CV also has a higher percentage in the lower depths of alternative read (R2 = 0749 ~ 0858). The use IQCM ensure a reduction of more than 100-fold in the cost per run compared to that achieved using a commercial approach defined reference materials.
Our common analytical performance NGS to detect CHIP and limitations are recognized as lower precision on the lower level of the load variants. This approach could also theoretically be extended to the general NGS assay for the detection of somatic variants. Given NGS reliable results and cost-effectiveness, we propose the use of NGS IQCM for QC tests in clinical laboratories.
Given the potential evidence of the real world (RWE) to inform understand the risk profile benefit from next-generation sequencing (NGS) based testing, we conducted a study to describe the landscape at this time whether and how the payer uses RWE as part of the decision-making scope and potential solutions to overcome barriers. We conducted a literature review of evidence scoping RWE existing framework to evaluate new technologies and identified barriers to clinical integration and evidence gaps for NGS. We synthesized the findings as a potential solution to improve the relevance and utility RWE for payer decision-making.

Description: Description of target: The protein encoded by this gene belongs to the pyridine nucleotide-disulfide oxidoreductase family, and is a member of the thioredoxin (Trx) system. Three thioredoxin reductase (TrxR) isozymes are found in mammals. TrxRs are selenocysteine-containing flavoenzymes, which reduce thioredoxins, as well as other substrates, and play a key role in redox homoeostasis. This gene encodes a mitochondrial form important for scavenging reactive oxygen species in mitochondria. It functions as a homodimer containing FAD, and selenocysteine (Sec) at the active site. Sec is encoded by UGA codon that normally signals translation termination.
